Troubleshooting: Count is outside the Optimum Counting Range

Modified on Mon, 23 Oct, 2023 at 4:18 AM

I receive the message "The count is below the optimum counting range".

This message appears in the results page when the cell concentration of the sample is below 5.0 x 104  cells/mL, which is outside the operating range of the cell counter. To resolve this, pellet the cell suspension by centrifugation and resuspend the cells in a smaller volume.

It could also be the case that the bulk sample solution has been left on the bench for some time, causing the cells to have gravitated towards the bottom of the vial. In this case, pipetting the solution up and down before loading the hemocytometer should result in a homogeneous cell suspension.

I receive the message "The count is above the optimum counting range".

This message appears in the results page when the cell concentration of the sample is above 1.0 x 107  cells/mL, which is outside the operating range of the cell counter. Dilute your cell suspension to obtain a lower concentration and count again. Learn more about the cell dilution calculation here: https://axionbio.freshdesk.com/a/solutions/articles/259739.
